    Association between prostate cancer development and TARBP1
    (Mongolian National University of Medical Sciences, 2025-03-31) ; ; ; ;
    Objective: Prostate cancer is the most frequent cancer among elderly men worldwide and second cause of cancer-related deaths in man. Prostate cancer characterized a clinically heterogeneous and genetic alterations in cells play a pivotal role in carcinogenesis. A detailed understandings of the genetic alterations in cancer cells are important for developing new therapeutic medicine. Transactivation response RNA-binding protein1 (TARBP1) encodes a protein with RNA-binding, tRNA methyltransferase activity, and RNA interference regulation functions, but its association with prostate cancer remains unclear. The aim of this study was to investigate clinical significance of TARBP1 in prostate cancer. Methods: Relative messenger ribonucleic acid (mRNA) and protein expression of TARBP1 in prostate cancer cell and tumor tissue was determined by qRT-PCR and Western blot analysis. LNCaP cells with lentivirus vector to silence the expression of TARBP1 were injected into nude mice. After 24 days, the mice were killed and tumor tissues were excised. The size of tumors was measured using a ruler and body weight of mice were weighed using a scale. Results: Through qRT-PCR and Western blot analysis, we validated that the expression levels of TARBP1 was decreased in TARBP1-knockdown prostate cancer cell compared with cells transfected with negative control LV-shNC RNA. The tumor growths were significant smaller in the group B (LNCaP-LV-shTARBP1) compared with group A (LNCaP-LV-shNC). Knocking down TARBP1 significantly suppressed the tumor progression of BALB/C nude xenograft. Moreover, body weight of mice in LNCaP-LV-shNC group increased significant slowly compared with LNCaP-LV-shTARBP1 group. The result of western blot analysis for TARBP1 in tumor tissues from xenograft nude mice bearing orthotopic LNCaP tumors shows that the expression of TARBP1 was also significantly decreased in the LNCaP-LV-shTARBP1 group compared with LNCaP-LV-shNC group (p < 0.05). Conclusion: We revealed that the TARBP1 highly expressed in prostate cancer and its role in promoting tumor growth. TARBP1 could serve as a potential therapeutic target for prostate cancer treatment and it is also a biomarker for tumor diagnosis and prognosis.

    Objective: Renal cell carcinoma (RCC) is one of the most common malignancies of the kidney, with genetic factors playing a critical role in its development. Among the key genetic factors, TP53 and MDM2 gene polymorphisms have been implicated in cancer susceptibility, but their association with RCC remains unclear in different populations. This study evaluated the association of TP53 and MDM2 gene polymorphisms with RCC risk among Mongolian individuals. Method: In a hospital-based case-control study, 88 RCC cases and 88 matched controls were analyzed for TP53 and MDM2 variants using RFLP-PCR based genotyping. Results: The TP53 CG and CC genotypes and MDM2 TG and GG genotypes were not significantly associated with RCC risk when considered individually. However, individuals carrying the MDM2 GG and TP53 CC genotypes showed elevated RCC risk, although statistical significance was not reached. No strong interaction effects between genotypes and clinical risk factors such as smoking or urinary tract disease were observed. Conclusion: While TP53 and MDM2 polymorphisms showed trends toward association with RCC, larger studies are warranted to clarify their roles in genetic susceptibility within the Mongolian population.

    Objective: The current investigation seeks to elucidate the regulatory impact of the long non-coding RNA GSEC (lncRNA GSEC) on oncogenic mechanisms in oral squamous cell carcinoma. Methods: The expression dynamics of GSEC were systematically mapped across in vitro systems, including healthy oral keratinocyte controls (NHOK) and their malignant counterparts (SCC-25, SCC-9, CAL-27), utilizing standardized qRT-PCR protocols. CAL-27 (highest GSEC expression) underwent siRNA-mediated knockdown. The assessment of cellular functions such as proliferation, apoptosis regulation, mobility, and invasion was conducted utilizing validated protocols: CCK-8 for cell viability assessment, flow cytometry for apoptotic cell identification, Transwell systems for measuring invasive ability, and scratch assay for migration analysis. mRNA and protein levels of proliferation/apoptosis markers (Ki67, PCNA, BCL-2, BAX, caspase-9) were analyzed by qRT-PCR, Western blot, and immunofluorescence. Results: GSEC was significantly upregulated in OSCC cells vs. NHOK (p<0.05), peaking in CAL-27 (p<0.05 vs. SCC-9/SCC-25). Downregulation of GSEC inhibited the proliferation, migration, and invasive capabilities of OSCC cells and promoted apoptosis with statistical significance (p<0.05). A decrease in Ki67 and PCNA expression, accompanied by an increase in BAX and caspase-9 and a decrease in BCL-2 levels, was detected at the mRNA and protein levels, with all reaching statistical significance at the p<0.05 threshold. Conclusion: GSEC overexpression promotes OSCC malignancy by driving proliferation and inhibiting apoptosis. Targeting GSEC may offer diagnostic and therapeutic potential for OSCC.

    Introduction: Prostate cancer is a prevalent malignancy among elderly men, with bioinformatics playing a crucial role in advancing diagnosis and treatment paradigms. Recent studies have highlighted the significance of nucleotide metabolism (NM) in Prostate cancer development and progression, linking it to aggressive cancer phenotypes characterized by uncontrolled proliferation and metastasis. Understanding NM-related genes (NMRGs) could provide insights into Prostate cancer pathogenesis and therapeutic targets.Methods: This paper analyzed TCGA-PRAD and GSE70769 datasets to identify critical modules associated with NMRGs using weighted gene co-expression network analysis (WGCNA). Differentially expressed genes (DEGs) between Prostate cancer and control samples were extracted from the TCGA-PRAD dataset, with overlaps identified as NM-related DEGs (DE-NMRGs). A biochemical recurrence (BCR)-free risk model was constructed from 396 Prostate cancer samples, and patients were classified into high- and low-risk groups based on median risk scores. A nomogram model integrating key prognostic factors was developed to predict BCR rates.Results: This paper identified 5 prognostic genes: RGS11, KAT2A, MXD3, TARBP1, and WFIKKN. The low-risk group exhibited significantly higher BCR-free survival rates, ESTIMATE scores, and immunophenoscore (IPS) scores compared to the high-risk group. Additionally, potential therapeutic agents, including KU-55933 and Wee1 inhibitors, were proposed. Conclusions: The identified prognostic genes present promising targets for Prostate cancer diagnosis and treatment, emphasizing their importance in predicting biochemical recurrence and tailoring personalized therapeutic strategies for patients.

    Association of GSTM1, NAT2 Gene Polymorphisms and Susceptibility to Renal cell carcinoma in Mongolia: A Case-Control Study
    OBJECTIVE: This study aimed to assess the impact of glutathione S-transferase M1 (GSTM1) and N-acetyltransferase 2 (NAT2) polymorphisms on renal cell carcinoma (RCC) risk in Mongolian individuals, both independently and in combination with smoking and urinary tract diseases (UTDs). METHODS: This hospital-based case-control study included 88 histologically confirmed RCC patients and 88 cancer-free controls, matched by age and sex. Genotyping of GSTM1 and NAT2 polymorphisms was performed using PCR-RFLP. RESULTS: There were 34 men and 54 women, with a mean age of 51.9 ± 13.2 years. The results revealed that NAT2 low acetylator genotype significantly increased risk of RCC (cOR=2.077, 95%CI=1.072-4.025, p=0.03). Notably, WT/M3 genotype was significantly associated with RCC risk (aOR=9.1, 95% CI=1.138-72.783; p=0.037). GSTM1-positive genotype significantly increased RCC risk when combined with NAT2 low acetylator genotypes (cOR=3.304, 95% CI=1.311-8.327, p=0.011). Among smokers, individuals with GSTM1-null genotype had an increased risk of RCC (cOR=4.654, 95% CI=1.458-14.86, p=0.009). Additionally, NAT2 low acetylator genotype significantly increased RCC risk in smokers (cOR=6.596, 95% CI=2.26-19.255, p=0.001). Furthermore, both GSTM1 and NAT2 genotypes were associated with significantly increased RCC risk following stratification by a history of urinary tract diseases. CONCLUSION: The findings suggest that the NAT2 WT/M3 polymorphism, along with smoking and UTDs, contributes to RCC susceptibility in Mongolian cases.
